Human MYH2 (Myosin-2) ELISA Kit

Assay Genie provides a Sandwich ELISA, Double Antibody kit for the quantitative measurement of MYH2 (Myosin-2), a skeletal muscle myosin heavy chain implicated in muscle contraction and cytoskeleton organisation, and linked to conditions such as inclusion body myopathy type 3. The assay is reactive with human samples, and is validated for use with serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples.

Performance characteristics quoted by Assay Genie include a sensitivity of 46.875 pg/mL and a working range of 78.125–5000 pg/mL. Recovery in spiked samples averages 93% from serum (range 87–105%), 94% from EDTA plasma (range 88–103%) and 95% from heparin plasma (range 85–102%). Linearity across 1:2, 1:4 and 1:8 dilutions is 87–98%, 82–90% and 81–97% for serum; 87–104%, 82–88% and 81–100% for EDTA plasma; and 87–95%, 88–98% and 85–99% for heparin plasma. Reagents should be stored at 2–8°C for up to 12 months, with standards and the coated plate additionally suitable for 12 months at -20°C.

The procedure follows a standard sandwich workflow: equilibrate reagents, load standards and samples on the pre-coated plate and incubate at 37°C for 90 minutes; add biotinylated detection antibody and incubate at 37°C for 60 minutes; add HRP-Streptavidin (SABC) and incubate at 37°C for 30 minutes; develop with TMB substrate in the dark for 10–20 minutes; then add stop solution and read absorbance at 450 nm. Components supplied include a dismountable ELISA microplate, lyophilised standard, biotin-labelled antibody concentrate, HRP-Streptavidin conjugate, TMB substrate, sample and antibody dilution buffers, SABC dilution buffer, stop solution, 25× wash buffer, plate sealer and a technical manual.
