Description

From Assay Genie, this Sandwich ELISA Kit is designed for the quantitative measurement of human Myelin-associated glycoprotein (MAG), also referred to by synonyms Siglec-4a and GMA. The assay is intended for use with serum, plasma, cell culture supernates, tissue homogenates and other biological fluids, and detects both natural and recombinant human MAG. The product is supplied for research use only.

MAG is a single-pass type I membrane glycoprotein belonging to the immunoglobulin superfamily SIGLEC family of sialic acid-binding Ig-like lectins, with synonyms also reported as S-MAG, Siglec-4 and SIGLEC4A. The protein localises to the cell membrane and membrane rafts, preferentially binds alpha-2,3-linked sialic acid and ganglioside Gt1b, and mediates sialic acid-dependent interactions between myelinating cells and neurons by binding neuronal gangliosides and the glycoproteins RTN4R and RTN4RL2. MAG exists as a monomer or homodimer, contributes to maintenance of normal axon myelination, protects motoneurons against apoptosis, and acts as a negative regulator of neurite outgrowth. Dysregulation of MAG has been associated with neurological disorders including multiple sclerosis and peripheral neuropathies, supporting its measurement as a research biomarker in studies of demyelinating disease.

Manufacturer-reported analytical specifications include a detection range of 0.156-10ng/mL with a sensitivity of 0.036ng/mL, an intra-assay CV of 5.3% and an inter-assay CV of 8.9%. Reported recovery in serum is 91-103% (average 97%) and in plasma 93-105% (average 99%). Linearity has been demonstrated across 1:2 to 1:16 serial dilutions of serum, EDTA plasma and heparin plasma samples. The UniProt accession for human MAG is P20916 with a reported molecular weight of 66,224 Da, and the NCBI Gene ID is 4099.
