Description

The Assay Genie sandwich ELISA format pairs a pre-coated capture antibody with a biotinylated detection antibody, allowing two independent epitopes on the MYC protein to be interrogated in each well. The microplate is supplied as 12 removable 8-well strips, and the kit ships with concentrated wash buffer, dilution buffer, standard diluent, TMB substrate, and stop solution. Researchers should bring all reagents to room temperature (~20–25 °C) before use, allow 30 minutes for equilibration, and protect the substrate from direct light. Samples in serum or plasma are typically applied directly or after a brief dilution in assay buffer; cell lysates may require homogenization in a non-denaturing buffer followed by centrifugation at 1000 x g for 10 minutes at 4 °C.

Because c-Myc is a basic helix-loop-helix transcription factor that regulates ribosomal biogenesis, glycolysis, mitochondrial function, and apoptosis, quantitative measurement supports studies of tumor biology, signal transduction downstream of Wnt, Notch, and MAPK pathways, and screens for compounds that perturb proliferation in cancer cell lines. The kit is configured for one full 96-well plate run, generating a seven-point standard curve in duplicate plus up to 40 unknown wells, with absorbance read at 450 nm and a 570 nm reference correction. Standard curves generally span ~31–2000 pg/mL, supporting detection of physiological and overexpressed MYC levels across human sample types in vitro.
