The MUSK protein is a transmembrane receptor tyrosine kinase essential for the formation and maintenance of the neuromuscular junction, where it is activated by the heparan sulfate proteoglycan agrin and signals through the cytoplasmic adaptor Dok-7 to cluster acetylcholine receptors on the postsynaptic membrane. Loss-of-function mutations in MUSK cause congenital myasthenic syndrome, and circulating autoantibodies against MUSK define a distinct seronegative subset of myasthenia gravis patients. Researchers use this Assay Genie kit to quantify MUSK levels in skeletal muscle homogenates, motor-neuron lysates, and cell culture supernatants. The assay uses paired capture and detection antibodies in a sandwich format that yields a colored readout on a standard microplate reader, requiring a small sample volume per well, typically around 100 µL. Each kit ships with a pre-coated plate, lyophilized standards, biotinylated detection reagent, streptavidin-HRP conjugate, wash buffer, substrate, and stop solution. Store sealed components refrigerated and protect reagents from light; avoid repeated freeze-thaw cycles of working solutions.
