Description

Assay Genie supplies a Sandwich ELISA kit designed for the quantitative measurement of Human Multidrug resistance-associated protein 1 (ABCC1). Synonyms listed include MRP1, ATP-binding cassette sub-family C member 1, Glutathione-S-conjugate-translocating ATPase ABCC1, Leukotriene C(4) transporter, and LTC4 transporter. The assay recognises natural and recombinant human ABCC1 in serum, plasma, tissue homogenates, cell culture supernates and other biological fluids, with a colorimetric detection method.

Stated performance characteristics are a detection range of 0.156-10ng/mL, a sensitivity of 0.082ng/mL, an intra-assay CV of 5.0%, and an inter-assay CV of 9.4%. Recovery in serum averages 97% with a range of 91-103%, and in plasma averages 99% with a range of 93-105%. Linearity at 1:2, 1:4, 1:8 and 1:16 dilutions is reported for serum (102-112%, 91-101%, 100-112%, 93-103%), EDTA plasma (103-114%, 112-121%, 93-105%, 85-95%) and heparin plasma (96-108%, 96-105%, 105-115%, 104-113%).

ABCC1 is a multi-pass cell membrane protein of the MRP subfamily of ABC transporters that mediates export of organic anions and drugs from the cytoplasm and ATP-dependent transport of glutathione and glutathione conjugates, leukotriene C4, estradiol-17-beta-o-glucuronide, methotrexate, antiviral drugs and other xenobiotics. It confers resistance to anticancer drugs by reducing drug accumulation via ATP- and GSH-dependent export, hydrolyses ATP with low efficiency, catalyses export of sphingosine 1-phosphate from mast cells, and participates in the inflammatory response through leukotriene C4 export. The kit is for research use only within the Signal Transduction research area, with UniProt accession P33527.
