Human Mucin-20/MUC20 ELISA Kit

This Assay Genie ELISA Kit is a colorimetric, double-antibody Sandwich ELISA designed for the quantitative measurement of Mucin-20 (MUC20) in human samples. The kit is reactive with human samples and validated for serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Reagents are stored at 2–8°C with a shelf life of 12 months under those conditions.

Linearity was demonstrated by serial dilution across sample matrices: serum ranged from 85–104% (1:2), 92–104% (1:4), and 94–105% (1:8); EDTA plasma from 83–98%, 83–93%, and 84–95%; and heparin plasma from 83–98%, 88–100%, and 89–100%. Spike-and-recovery testing showed serum recovery of 89–104% (average 99%), EDTA plasma of 95–102% (average 98%), and heparin plasma of 86–100% (average 96%).

The assay workflow includes equilibrating reagents and TMB substrate to room temperature, loading standards, samples, and controls on the pre-coated plate for a 90-minute primary incubation at 37°C, biotin-labeled detection antibody incubation at 37°C for 60 minutes, HRP-Streptavidin (SABC) binding at 37°C for 30 minutes, TMB substrate color development in the dark for 10–20 minutes, and immediate absorbance measurement at 450 nm following addition of stop solution. Serum is collected after clotting and centrifuged at 1000 × g for 20 minutes, while plasma is collected in anticoagulant tubes and centrifuged at 1000 × g for 15 minutes at 2–8°C.
