Description

A high-quality sandwich ELISA kit from Assay Genie for the quantitative measurement of human Mucin 17 (MUC17). The assay uses a double-antibody sandwich format in which a capture antibody is pre-coated onto the plate and a biotin-conjugated antibody serves as the detector, with HRP-Streptavidin and TMB substrate used for colorimetric readout at 450 nm. The kit has a sensitivity of 9.375 pg/mL and a detection range of 15.625-1000 pg/mL. Reactivity is human and the UniProt reference for the target is Q685J3. Validated sample types include serum, plasma, cell culture supernatant, cell or tissue lysate and other liquid samples.

MUC17 is a mucin protein that probably plays a role in maintaining homeostasis on mucosal surfaces. Dysregulation of MUC17 expression has been associated with various diseases, including gastrointestinal cancers and inflammatory bowel disease, making it a relevant biomarker in mucosal biology and disease research. Reported linearity for serum (n=5) across 1:2, 1:4 and 1:8 dilutions is 88-104%, 87-105% and 91-102%, respectively; for EDTA plasma (n=5) 85-103%, 84-95% and 83-101%; and for heparin plasma (n=5) 87-100%, 82-94% and 85-96%. Recovery ranges across the same sample types fall within 88-99% (serum), 88-100% (EDTA plasma) and 88-99% (heparin plasma) with corresponding averages of 93%, 95% and 95%. The kit is supplied with a pre-coated microplate, lyophilized standard, biotin-labelled detection antibody, HRP-Streptavidin conjugate (SABC), TMB substrate, sample and antibody dilution buffers, wash buffer (25X), stop solution and plate sealer, with most liquid reagents stored at 2-8°C.

The quick protocol involves incubating standards or samples for 90 minutes at 37°C, adding biotin-labelled antibody for 60 minutes at 37°C, adding SABC working solution for 30 minutes at 37°C, developing with TMB substrate for 10-20 minutes and reading absorbance at 450 nm after adding stop solution. For research use only.
