Human Mucin-12/MUC12 ELISA Kit

A Sandwich ELISA, Double Antibody assay from Assay Genie designed for the precise measurement of Mucin-12 (MUC12) in human biological samples. The kit is intended for research use and is suited to serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. MUC12 is a mucin protein known for its role in mucosal immunity and maintaining epithelial barrier function, with abnormalities in its expression linked to inflammatory bowel disease, asthma, and other gastrointestinal disorders, making it a valuable biomarker for studying these conditions and developing potential treatments.

Reported assay performance includes a sensitivity of 0.375 ng/mL and a range of 0.625–40 ng/mL. Linearity across 1:2, 1:4 and 1:8 dilutions (n = 5) is reported as 87–102%, 90–105% and 87–97% for serum; 88–101%, 82–100% and 84–98% for EDTA plasma; and 84–99%, 83–98% and 87–93% for heparin plasma. Spike-and-recovery values (n = 5) are given as 85–105% (average 96%) for serum, 87–104% (average 98%) for EDTA plasma, and 91–105% (average 96%) for heparin plasma. The protocol proceeds through reagent and plate preparation, primary incubation at 37°C for 90 minutes, biotin-labelled detection antibody binding at 37°C for 60 minutes, HRP-Streptavidin binding at 37°C for 30 minutes, TMB colour development in the dark for 10–20 minutes, and a stop reaction read at 450 nm.

Sample preparation guidance is provided for serum, plasma, tissue homogenate, cell culture supernatant, cell lysate and other sample types. The kit components include a dismountable ELISA microplate, lyophilised standard, biotin-labelled antibody (100X), HRP-Streptavidin conjugate SABC (100X), TMB substrate, sample dilution buffer, antibody dilution buffer, SABC dilution buffer, stop solution, 25X wash buffer, plate sealer and a technical manual. Storage is at 2–8°C for 12 months, with specified storage conditions for individual components.
