Description

From Assay Genie, this Competitive ELISA Kit (antigen-coated, colorimetric detection) is designed for the quantitative measurement of MTPAP, the human Poly (A) RNA polymerase, in human samples. In the assay, analyte in the sample or standard competes with antigen pre-coated on the microplate for binding sites on a biotinylated detection antibody, with HRP-Streptavidin (SABC) and TMB substrate used for signal generation; absorbance is read at 450 nm.

The kit is reactive with human samples and has been validated for serum, plasma, cell culture supernatant, cell or tissue lysates, and other biological fluids. Reported sensitivity is 0.188 ng/mL with a measuring range of 0.313-20 ng/mL. Kits are shipped on ice packs and stored at 2-8°C (with selected components at -20°C for longer-term storage). The product is intended for Research Use Only. MTPAP is an enzyme involved in polyadenylation, a step in mRNA processing, and its dysregulation has been linked to neurodegenerative disorders and mitochondrial dysfunction.

Validated linearity and recovery data are provided for serum, EDTA plasma, and heparin plasma samples (n=10 each), with reported recovery ranges of 89-99% (average 94%) for serum, 89-98% (average 92%) for EDTA plasma, and 88-105% (average 96%) for heparin plasma. The standard procedure involves pre-washing the plate, adding 50 µL of standard or sample together with 50 µL of biotin-labelled antibody per well, incubating at 37°C, washing, adding SABC working solution, developing with TMB substrate, stopping the reaction, and reading absorbance at 450 nm. Known kit aliases include PAPD1, PAP, TUTase 1, mtPAP, Terminal uridylyltransferase 1, PAP-associated domain-containing protein 1, and Polynucleotide adenylyltransferase (Uniprot Q9NVV4).
