Human MTF1/Metal regulatory transcription factor 1 ELISA Kit

From Assay Genie, this sandwich ELISA kit employs a double-antibody format for the measurement of human MTF1 (Metal Regulatory Transcription Factor 1), a transcription factor that regulates expression of metal-responsive genes and contributes to cellular metal homeostasis. The assay is validated for use with human serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples.

Performance parameters include a sensitivity of 0.469 ng/mL and a detection range of 0.781-50 ng/mL, with colorimetric detection. Storage is at 2-8°C for 12 months. Validation data showed linearity in serum at dilutions 1:2, 1:4 and 1:8 of 93-105%, 86-105% and 86-103%; in EDTA plasma of 89-101%, 82-88% and 89-100%; and in heparin plasma of 81-86%, 81-93% and 84-98%. Mean recovery was 96% for serum (range 94-100%), 90% for EDTA plasma (range 86-97%) and 95% for heparin plasma (range 86-105%).

The workflow covers reagent equilibration, plate loading of standards and samples, primary incubation at 37°C for 90 minutes, biotinylated detection antibody incubation at 37°C for 60 minutes, HRP-streptavidin binding for 30 minutes, TMB development in the dark for 10-20 minutes, and absorbance measurement at 450 nm. Sample preparation protocols describe centrifugation at 1000 × g for 20 minutes for serum, 1000 × g for 15 minutes at 2-8°C for plasma, homogenisation with protease inhibitors for tissue, centrifugation at 2500 rpm for 5 minutes for cell culture supernatant, and lysis buffer with protease inhibitors for cell lysates.
