MSK1, also known as Mitogen- and Stress-activated protein Kinase 1 and encoded by the RPS6KA5 gene, is a nuclear serine/threonine kinase of approximately 88 kDa that contains two kinase domains within a single polypeptide chain. It is activated downstream of the ERK1/2 and p38 MAPK cascades and phosphorylates substrates that include the transcription factors CREB and ATF1, as well as chromatin components such as histone H3. Through these activities MSK1 couples stress and mitogenic signals to immediate-early gene transcription and chromatin remodeling, and is studied in research on inflammatory signaling, immune regulation, and tumor cell biology.

This Assay Genie kit is configured to detect human MSK1 quantitatively, employing a human-specific antibody pair to selectively capture the target analyte. The plate-based assay produces an absorbance-based readout for use with standard microplate readers, and each kit contains a pre-coated microplate, lyophilized standards, detection antibody, substrate solution, wash buffer, and protocol sufficient to evaluate a full plate of samples. The kit is intended for research use only and is not cleared for clinical or diagnostic applications.

Storage and handling: keep kit components refrigerated upon arrival and protect chromogen and substrate vials from prolonged light exposure. Allow all reagents to equilibrate to room temperature before use, and aliquot reconstituted standards and prepared samples to minimize freeze-thaw cycles. Plate layout, standard curve preparation, and recommended sample dilution guidelines are detailed in the enclosed product protocol.
