Description

From Assay Genie, this sandwich ELISA kit is designed for the precise quantification of human MRP9 (ABCC12), a member of the ATP-binding cassette (ABC) transporter superfamily implicated in drug resistance and cellular detoxification, with dysregulation linked to cancer, cystic fibrosis and neurodegenerative disorders. The double-antibody sandwich format uses colorimetric detection and is intended for use with serum, plasma, cell culture supernatants, cell or tissue lysates and other liquid samples of human origin.

Assay performance specifications include a sensitivity of 0.188 ng/mL and a working range of 0.313–20 ng/mL. Spiking recovery was 88–95% (average 91%) in serum, 86–97% (average 92%) in EDTA plasma and 85–103% (average 93%) in heparin plasma (n = 5 per matrix). Linearity across 1:2, 1:4 and 1:8 dilutions was 87–105% / 85–104% / 87–105% for serum, 84–100% / 89–101% / 88–95% for EDTA plasma, and 86–99% / 89–100% / 88–97% for heparin plasma (n = 5). Unopened kits are stored at 2–8°C for 12 months.

The workflow comprises a 90-minute primary incubation at 37 °C, a 60-minute biotin-labelled detection antibody step, 30 minutes with HRP-Streptavidin (SABC), 10–20 minutes of TMB colour development in the dark, and absorbance reading at 450 nm after addition of stop solution. Sample handling notes from the manufacturer include allowing blood to clot and centrifuging serum at 1000 × g for 20 minutes, collecting plasma at 1000 × g for 15 minutes at 2–8 °C, and lysing cells with a protease-inhibitor-containing buffer before collection of the protein supernatant.
