Biological Target and Assay Specifications

The Assay Genie human MRC1 kit quantifies the macrophage mannose receptor (UniProt P22897, gene ID 4360), also designated CD206, MMR, CLEC13D, CLEC13DL, or MRC1L1. The protein is a ~175 kDa type I transmembrane C-type lectin that binds terminal mannose, fucose, and N-acetylglucosamine residues through its carbohydrate-recognition domains, mediating endocytosis of glycoproteins, lysosomal hydrolases, and microbial ligands.

Performance Characteristics

  • Standard curve range: typically 0.156-10 ng/mL, generated from recombinant human CD206.
  • Reported intra-assay CV: approximately 4.4%; inter-assay CV: approximately 7.9%.
  • Sample volume: 100 µL per well; total runtime around 3.5 hours at room temperature or overnight at 4 degrees C.
  • Read at 450 nm with optional 540 nm or 570 nm reference wavelength for plate correction.
  • Quantitative, sandwich-based colorimetric format supplied as eight removable 12-well strip plates.

Sample Preparation Guidance

  • Serum: clot 30 min at room temperature, centrifuge 10 min at 1000 x g, aliquot, and freeze at -80 degrees C; avoid repeated freeze-thaw cycles.
  • Plasma (EDTA or heparin): centrifuge 15 min at 1000 x g at 4 degrees C within 30 minutes of collection.
  • Cell culture supernatant: centrifuge 20 min at 1500 rpm at 4 degrees C before the assay.
  • Tissue homogenates: lyse in PBS with protease inhibitors, sonicate, then centrifuge 5 min at 5000 x g.
  • Cell lysates: solubilize in lysis buffer on ice, centrifuge 5 min at 14,000 x g, and quantify total protein prior to loading.

Research Context

CD206 is a widely cited marker of alternatively activated M2 macrophages, and circulating or tissue levels are studied in tumor immunology, fibrotic disease, and chronic inflammatory models. Quantifying soluble receptor shed from macrophage surfaces supports investigations into M1/M2 polarization kinetics, tumor-associated macrophage profiling in solid tumors, and outcomes of in vitro polarization assays using IL-4, IL-10, or IL-13 stimulation of peripheral blood monocytes or THP-1-derived macrophages.
