The Human MPPE1 ELISA Kit from Assay Genie is a quantitative enzyme-linked immunosorbent assay intended for the in vitro measurement of Metallophosphoesterase 1 (also known as PGAP5 or Post-GPI Attachment to Proteins 5) in human-derived samples. MPPE1 is a Golgi-resident enzyme involved in the fatty acid remodeling of glycosylphosphatidylinositol (GPI) lipid anchors, a process essential for the proper membrane attachment of many cell-surface proteins. Variants in the MPPE1 gene have been associated with hyperphosphatasia with mental retardation syndrome, making accurate quantification of this protein relevant to studies of glycosylphosphatidylinositol anchor biosynthesis and related neurodevelopmental disorders.

The kit is supplied as a 96 Assays microplate with all reagents required to perform the assay, including pre-coated wells, lyophilized standards, detection antibody, and substrate solution. The assay follows a two-antibody ELISA workflow with absorbance readout, providing a standard curve-based quantification of endogenous MPPE1 protein. A sample volume of 100 µL is loaded per well, and the lyophilized standard is reconstituted to generate a serial dilution series for interpolation. The assay typically requires 2-4 hours of incubation time with minimal hands-on steps, making it suitable for routine research laboratory workflows.

For research use only. Store kit components refrigerated, and avoid repeated freeze-thaw cycles to preserve reagent activity. The capture antibody demonstrates human-specific reactivity with no significant cross-reactivity with homologous proteins from rat or mouse, ensuring accurate measurement in human-derived matrices.
