Human MPO ELISA Kit

Designed by Assay Genie for the quantitative measurement of human myeloperoxidase (MPO), this sandwich (double antibody) ELISA supports in vitro determination of MPO concentrations in serum, plasma, cell culture supernatant, cell or tissue lysate, and other biological fluids. The colorimetric assay has a measurement range of 1.56–100 ng/ml and a sensitivity of 0.938 ng/ml.

The workflow uses an antibody pre-coated onto a microplate; samples and standards are added, followed by a biotinylated detection antibody and HRP-conjugated streptavidin. TMB substrate produces a colorimetric reaction read at 450 nm, with the OD value proportional to analyte concentration. The kit also includes a validated Bradford Reagent to quantify total protein for sample normalization. Reported performance characteristics include intra-assay CV <8%, inter-assay CV <10%, and average spike-recovery rates of 93% in serum, 100% in EDTA plasma, and 95% in UFH plasma. Components are stored at 2–8°C (with selected items stored at -20°C), and the kit is intended for research use only.

For tissue homogenates, buffers containing NP-40, Triton X-100, or DTT should be avoided as they strongly inhibit the assay. Tissues with high endogenous peroxidase activity, such as liver, kidney, or pancreas, may require pre-treatment with 1% H₂O₂ for 15 minutes before testing to minimise potential interference with the chromogenic substrate.
