Phosphotyrosine STAT5 detection provides a direct readout of JAK-STAT pathway activation, capturing only the activated, tyrosine-phosphorylated form of STAT5 rather than total protein levels. The Assay Genie Phosphorylation ELISA kit uses a microplate-based protocol compatible with cell lysates and tissue lysates, making it well suited for cytokine, growth factor, and oncogenic signaling studies where STAT5 is phosphorylated downstream of receptors for IL-2, IL-3, IL-7, IL-15, GM-CSF, erythropoietin, thrombopoietin, growth hormone, and prolactin.

Validated for use with human samples, the assay also cross-reacts with mouse and rat STAT5, allowing translational researchers to compare activated STAT5 levels across species in matched cell line panels and primary tissue specimens. The kit contains a pre-coated plate in a 96 assays configuration with all reagents required to run a full plate of samples and standards in parallel, including capture and detection antibodies specific for the phosphotyrosine epitope. Store the unopened kit refrigerated at 4 °C; protect the conjugated detection antibody from light and avoid repeated freeze-thaw cycles of the standard to preserve calibration accuracy. Reconstituted standards should be aliquoted and frozen for longer term storage.

This kit supports research into hematological malignancies, immune cell activation, T cell and NK cell signaling, mammary gland development, and metabolic regulation. Researchers investigating JAK2 V617F or FLT3-ITD driven leukemias, where constitutive STAT5 phosphorylation is a hallmark, can use this assay to quantify pathway activation in treated versus untreated cells. The supplied protocol recommends preparing lysates in RIPA or equivalent buffer with freshly added phosphatase inhibitors to preserve the phosphorylation mark; results are read on a standard microplate reader after the color development step.
