SHP2 (encoded by PTPN11) is a non-receptor protein tyrosine phosphatase whose catalytic function is modulated by phosphorylation at residue Y542, a site within its N-terminal SH2 domain that reflects upstream receptor tyrosine kinase activation. This dual-analyte ELISA from Assay Genie measures both phospho-SHP2 (Y542) and total SHP2 protein from the same lysate preparation, allowing researchers to express phospho-SHP2 levels normalized to total SHP2 abundance and thereby control for differences in sample input or expression across experimental conditions.

Validated with cell lysates and tissue lysates, the assay produces a colorimetric read-out on standard microplate readers. It is part of the PharmaGenie product line and supports research use only in growth factor and cytokine signaling studies, characterization of oncogenic PTPN11 mutations implicated in juvenile myelomonocytic leukemia and Noonan syndrome models, validation of upstream receptor tyrosine kinase pathway components, and CRISPR or RNAi knockdown experiments.

Common applications include monitoring SHP2 pathway engagement after treatment with small-molecule SHP2 inhibitors, comparing phospho-SHP2 levels between tumor and matched normal tissue, profiling pathway reactivation kinetics following drug washout, and confirming target modulation in preclinical drug discovery programs directed at the RAS/MAPK and PI3K/AKT cascades downstream of SHP2.
