For researchers studying p53-mediated DNA damage signaling, the Assay Genie Phospho-P53 (S15) and Total P53 ELISA Kit delivers simultaneous measurement of phosphorylated p53 at serine 15 alongside total p53 protein. Phosphorylation at S15 is a hallmark event executed by ATM and ATR kinases during the cellular response to genotoxic stress, stabilizing p53 and triggering transcription of genes controlling cell cycle arrest and apoptosis. Profiling both the activated and total pools enables proper normalization when changes in overall p53 expression might otherwise confound interpretation of pathway activation.

This dual readout supports studies of DNA damage induction, tumor suppressor pathway dynamics, chemotherapy response, and preclinical compound screening across human, mouse, and rat samples. The kit accommodates cell lysates and tissue lysates, and is supplied in a 96 assays plate configuration compatible with standard microplate readers. For accurate quantitation, lysates should be prepared with phosphatase inhibitors to preserve the phospho-S15 epitope, and protein loading normalized across samples.

Assay Genie develops the PharmaGenie line for translational and preclinical research applications, and this kit is widely used to interrogate p53 pathway activity in oncology, toxicology, and DNA repair studies. The workflow fits established ELISA protocols and pairs effectively with western blot validation when orthogonal confirmation is desired.
