The Phospho-JNK (T183/Y185) PharmaGenie ELISA from Assay Genie uses two phosphorylation-site-specific antibodies that selectively capture JNK1, JNK2, and JNK3 only when both threonine 183 and tyrosine 185 within the activation loop are phosphorylated. Because these residues are conserved between the human, mouse, and rat orthologs, the same plate can be used to compare stress-kinase activation across cell lysates and tissue lysates from all three species without re-optimizing the antibody pair, giving true cross-reactivity for translational studies.

The assay is built for signaling research rather than clinical diagnostics, and is most often deployed to track JNK pathway engagement after cellular stress stimuli such as UV irradiation, osmotic shock, oxidative stress, or treatment with pro-inflammatory cytokines such as TNF-alpha and IL-1. Pairing the phospho-JNK readout with a total-JNK ELISA on parallel lysates lets researchers quantify pathway activation as a phospho/total ratio, the standard way to report JNK signaling in peer-reviewed publications.

Working on a standard microplate layout with a small sample volume of around 100 uL per well, the kit fits existing plate-reader workflows and uses standard curve-fitting on diluted recombinant phospho-JNK standards. Reagents are shipped on refrigerated cold packs; the kit components should be stored at 2-8 degrees C for short-term use, while capture and detection antibodies are kept at -20 degrees C for longer-term stability to preserve phospho-epitope binding activity.
