This dual-analyte immunoassay from Assay Genie quantifies both the active, dual-phosphorylated form of ERK2 at threonine 185 and tyrosine 187 and total ERK2 protein from human, mouse, and rat samples in a single microplate run. Detecting the activated kinase alongside the total protein pool enables researchers to compute a phospho-to-total ERK2 activation ratio, a standard metric for MAPK/ERK pathway engagement following growth factor, cytokine, or pharmacological stimulation.

ERK2 (MAPK1) is a 42 kDa serine/threonine kinase whose full catalytic activity depends on MEK1/2-mediated phosphorylation at the conserved T-E-Y motif. The kit is validated for cell lysates and tissue lysates prepared with standard RIPA or equivalent detergent buffers supplemented with protease and phosphatase inhibitor cocktails. Compatible research applications include cancer cell line profiling, neuronal signaling studies, immune-cell activation assays, and small-molecule MEK or Raf inhibitor screening, where distinguishing basal from stimulated ERK2 phosphorylation is critical.
