Description

From Assay Genie, this colorimetric Sandwich ELISA kit in a Double Antibody format is intended for the quantitative measurement of Motilin in human serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The workflow begins with equilibrating reagents and TMB substrate to room temperature, loading standards, samples and controls onto the pre-coated plate, and incubating at 37°C for 90 minutes to allow antigen binding. After a biotin-labeled detection antibody incubation at 37°C for 60 minutes and an HRP-Streptavidin (SABC) step at 37°C for 30 minutes, TMB is added and developed in the dark for 10–20 minutes; the reaction is then stopped and absorbance is read at 450 nm. The kit is supplied with reagents for storage at 2-8°C for up to 12 months.

Reported performance specifications include a sensitivity of 18.75 pg/mL and an assay range of 31.25-2000 pg/mL. Linearity on spiked samples is reported as 86-104% at 1:2, 94-105% at 1:4 and 90-100% at 1:8 for serum (n=5); 82-99%, 84-95% and 84-101% for EDTA plasma (n=5); and 90-98%, 80-96% and 81-99% for heparin plasma (n=5). Recovery ranges across the same matrices are 93-103% (average 96%) for serum, 88-105% (average 95%) for EDTA plasma and 86-105% (average 93%) for heparin plasma.

Motilin is a hormone that regulates gastrointestinal motility by stimulating smooth-muscle contraction in the digestive tract. Imbalances in motilin levels have been linked to digestive disorders such as dyspepsia and gastroparesis, positioning it as a useful biomarker for gastrointestinal research.
