Description

An ELISA from Assay Genie for the quantitative, colorimetric measurement of Myelin oligodendrocyte glycoprotein (MOG) in human samples. The kit uses a double-antibody sandwich format with a capture antibody pre-coated on the microplate, and absorbance is read at 450 nm after TMB-based color development. Compatible sample types include serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples.

Standards and appropriately diluted samples are incubated at 37°C for 90 minutes to allow the target analyte to bind the immobilized antibody. A biotinylated detection antibody specific to a separate epitope of the target protein is then added and incubated at 37°C for 60 minutes, forming a sandwich complex. After washing, HRP-conjugated Streptavidin is added for 30 minutes at 37°C, followed by TMB substrate incubated in the dark for 10–20 minutes. The reaction is stopped with an acidic solution and absorbance is measured at 450 nm; signal intensity is directly proportional to analyte concentration and is determined from a standard curve.

Linearity was evaluated by serial dilution (n=5 per matrix): at 1:2, 1:4 and 1:8 dilutions, serum was 89–98%, 90–105% and 86–105%; EDTA plasma was 84–95%, 82–101% and 89–98%; heparin plasma was 87–97%, 85–97% and 84–99%. Spike-and-recovery averaged 96% for serum (range 88–104%), 92% for EDTA plasma (range 88–100%) and 95% for heparin plasma (range 86–103%). Kits should be stored at 2–8°C for 12 months.
