Description

A Sandwich ELISA Kit from Assay Genie designed for the quantitative measurement of human MMP1 in biological samples. The assay uses a double-antibody sandwich ELISA format with colorimetric detection, with absorbance read at 450 nm, and provides a sensitivity of 46.875 pg/mL across a standard range of 78.125–5000 pg/mL. It is reactive with human samples and is suitable for use with serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid sample types.

The protocol involves equilibrating reagents and TMB substrate to room temperature, setting standard, test sample and zero control wells on the pre-coated plate, incubating at 37°C for 90 minutes to allow antigen binding, adding a biotin-labelled detection antibody with a further 60-minute incubation, adding HRP-Streptavidin (SABC) for 30 minutes, developing colour with TMB substrate in the dark for 10–20 minutes, then adding stop solution and reading absorbance at 450 nm. Sample preparation guidance covers serum (allow to clot, centrifuge at 1000 × g for 20 minutes and collect supernatant), plasma (anticoagulant tubes, 1000 × g for 15 minutes at 2–8°C), tissue homogenate (homogenise in PBS with protease inhibitors, centrifuge and collect supernatant), cell culture supernatant (2500 rpm for 5 minutes) and cell lysates.

Components supplied include a dismountable pre-coated microplate, lyophilised standard, biotin-labelled detection antibody concentrate, HRP-Streptavidin conjugate concentrate (SABC), TMB substrate, sample dilution buffer, antibody dilution buffer, SABC dilution buffer, stop solution, wash buffer, plate sealers and a technical manual. Reagents should be stored at 2–8°C and protected from direct light where indicated, with the microplate and standard also suitable for longer-term storage at –20°C. MMP1 (matrix metalloproteinase-1) is a key enzyme in extracellular matrix breakdown relevant to tissue remodelling research.
