Human MMP 9 ELISA Kit

Manufactured by Assay Genie, this sandwich ELISA kit is designed for the precise quantification of human Matrix Metalloproteinase-9 (MMP-9) in serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. MMP-9 is a key enzyme involved in the breakdown of extracellular matrix proteins and plays a crucial role in tissue remodeling, inflammation, and cell migration. Dysregulation of MMP-9 has been implicated in pathological conditions such as cancer, arthritis, and cardiovascular diseases, making it an important biomarker for disease progression and treatment monitoring.

The assay uses a double-antibody sandwich format with a colorimetric readout, delivering a sensitivity of 0.188 ng/mL and a measurable range of 0.313–20 ng/mL. Storage is recommended at 2–8°C for 12 months. The workflow includes reagent equilibration and plate setup, primary incubation at 37°C for 90 minutes, biotin-labeled detection antibody incubation for 60 minutes, HRP-streptavidin binding for 30 minutes, TMB substrate development in the dark for 10–20 minutes, and a stop reaction read at 450 nm. Performance is supported by linearity of 83–105% across serum, EDTA plasma, and heparin plasma at 1:2, 1:4, and 1:8 dilutions, with recovery ranging from 87–105% in the same matrices.

Sample preparation guidance covers serum clotting and centrifugation at 1,000 × g for 20 minutes, plasma collection with anticoagulant and centrifugation at 1,000 × g for 15 minutes at 2–8°C, tissue homogenization in PBS with protease inhibitors, cell culture supernatant clarification at 2,500 rpm for 5 minutes, and cell lysis with protease-inhibitor buffer.
