MLKL (mixed lineage kinase domain-like) is the terminal effector of necroptosis, a regulated form of inflammatory cell death. Upon phosphorylation by RIPK3 at Thr357 and Ser358, this ~54 kDa pseudokinase oligomerizes and translocates to the plasma membrane, where it disrupts membrane integrity and releases damage-associated molecular patterns. Measuring circulating MLKL concentrations therefore supports investigations into necroptosis-related conditions, including acute kidney injury, ischemic stroke, sepsis, inflammatory bowel disease, and multiple cancer settings.
