Description

The Assay Genie Human Mitogen-activated protein kinase 3 (MAPK3) ELISA Kit is a sandwich-format immunoassay developed for the quantitative measurement of MAPK3 in human samples. The assay uses a pre-coated microplate together with a biotinylated detection antibody and an Avidin-Horseradish Peroxidase (HRP) conjugate, with the enzyme–substrate reaction terminated by a sulphuric acid solution and the optical density read at 450 nm ± 2 nm. It is intended for research use only.

Reported performance characteristics include a detection range of 0.156-10ng/mL and a sensitivity of 0.067ng/mL, with an intra-assay CV of 4.3% and an inter-assay CV of 6.8%. Linearity has been evaluated in serum (e.g. 97-107% at 1:2 and 109-119% at 1:4), EDTA plasma (83-95% at 1:2; 104-113% at 1:4) and heparin plasma (108-117% at 1:2 and 1:4). Spike-and-recovery data show averages of 88% (range 82-94%) in serum and 90% (range 84-96%) in plasma. Validated sample types include serum, plasma, cell culture supernatant, tissue homogenates, saliva, urine, blood and other biological fluids, and the kit is compatible with routine laboratory formats.

MAPK3, also known as ERK1, p44-ERK1, ERK-1, p44-MAPK, MAP kinase 3, ERT2, PRKM3, Microtubule-associated protein 2 kinase, MAP kinase isoform p44, Insulin-stimulated MAP2 kinase, or Extracellular signal-regulated kinase 1 (Uniprot P27361), is a serine/threonine kinase that acts as an essential component of the MAP kinase signal transduction pathway, regulating processes such as cell growth, adhesion, survival and differentiation. The protein localises to the cytoplasm and nucleus, with autophosphorylation at Thr-207 promoting nuclear localisation, and the listed research area is Stem Cells. The assay plate, standard and detection reagents should be stored at -20°C, with the substrate kept at 4°C.
