The MITF ELISA from Assay Genie targets human microphthalmia-associated transcription factor, a basic helix-loop-helix leucine zipper protein that acts as the master regulator of melanocyte lineage commitment and is widely investigated in pigmentation biology, ocular development, and melanoma research. By quantitatively capturing MITF protein in experimental samples, the assay supports studies of melanogenesis pathways, MITF-M and MITF-A isoform expression, and downstream targets such as TYR, TYRP1, DCT, and PMEL, while also enabling investigation of MITF dysregulation in uveal and cutaneous melanoma cell lines and in patient-derived tumor specimens.

Supplied as a microplate kit with an absorbance-based readout, the assay is configured for use with tissue homogenates and cell lysates prepared from human samples, making it suitable for cultured melanocyte and melanoma cell models as well as dissected ocular and dermal tissue. Each plate provides 96 individual determinations, and the protocol follows a matched antibody pair workflow that researchers can run on standard plate readers commonly available in biochemistry and dermatology laboratories.

Typical applications include pathway analysis following BRAF or NRAS perturbation, characterization of MITF expression in response to cAMP, UV, or alpha-MSH stimulation, screening of compounds that modulate pigmentation, and validation of transcriptomic findings at the protein level in melanoma research. The kit is intended for research use only and is not intended for diagnostic procedures.
