Description

This competitive ELISA kit from Assay Genie is designed for the quantitative measurement of metanephrine in human samples. Metanephrine is a metabolite of the catecholamines epinephrine and norepinephrine, which are involved in the body's stress response and blood pressure regulation. The assay uses a microplate pre-coated with antigen and a biotin-labelled antibody, with signal developed using TMB substrate, stopped with sulphuric acid, and absorbance read at 450 nm.

Reactivity is with human samples, and the kit is validated for use with serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The assay has a sensitivity of 9.375 pg/mL and a working range of 15.625-1000 pg/mL. Linearity has been characterised: serum (n=10) at 1:2, 1:4 and 1:8 dilutions gives 88-101%, 88-97% and 85-92%; EDTA plasma (n=10) gives 86-100%, 85-95% and 83-98%; heparin plasma (n=10) gives 87-102%, 83-97% and 85-100%. Recovery ranges from 88-103% (average 97%) for serum, 88-103% (average 96%) for EDTA plasma, and 86-104% (average 93%) for heparin plasma.

The protocol involves adding 50 µL of standard or sample together with 50 µL of biotin-labelled antibody per well, incubating at 37°C for 45 minutes, washing, adding HRP-streptavidin conjugate, developing with TMB substrate, and reading at 450 nm. Reagents are stored at 2-8°C (sealed) and must not be cryopreserved. The kit supports research into conditions associated with catecholamine excess, including pheochromocytoma, with applications in endocrinology, oncology and cardiovascular medicine, and is intended for research use only.
