Human Metadherin/MTDH ELISA Kit

Assay Genie's Human Metadherin (MTDH) ELISA Kit is a colorimetric Sandwich ELISA built on a double-antibody format for the quantitative measurement of MTDH in human samples. Accepted sample types include serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples, with all reagents stored at 2-8°C for 12 months.

Performance characteristics include a sensitivity of 0.938 pmol/mL and an assay range of 1.563-100 pmol/mL. Linearity was demonstrated at 1:2, 1:4 and 1:8 dilutions for serum (87-101%, 86-105%, 85-99%), EDTA plasma (85-100%, 82-101%, 83-101%) and heparin plasma (90-99%, 83-98%, 82-98%), each tested with n=5. Recovery averaged 94% in serum (range 85-103%), 94% in EDTA plasma (range 86-100%) and 97% in heparin plasma (range 90-105%).

MTDH, also referred to as astrocyte elevated gene-1 (AEG-1), is involved in cell proliferation, migration and invasion, and its dysregulation has been linked to cancer, inflammatory disorders and metabolic diseases. The workflow uses primary incubation at 37°C for 90 minutes, biotin-labelled detection antibody for 60 minutes, HRP-Streptavidin (SABC) for 30 minutes, TMB colour development in the dark for 10-20 minutes, and absorbance measurement at 450 nm after adding stop solution.
