Metadherin (MTDH), also known as AEG-1 (Astrocyte Elevated Gene-1) or LYRIC, is a multifunctional oncoprotein anchored at the plasma membrane with additional nuclear and endoplasmic reticulum localization. MTDH drives tumor initiation, metastatic dissemination, and chemoresistance across hepatocellular carcinoma, breast cancer, glioblastoma, and prostate cancer, while activating PI3K/Akt, Wnt/β-catenin, and NF-κB signaling cascades.
