Human MCP4 protein/CCL13 ELISA Kit

A Sandwich ELISA Kit from Assay Genie, this double-antibody immunoassay is designed for the quantitative detection of human MCP4 (CCL13) in serum, plasma, cell culture supernatant, cell or tissue lysates, and other liquid samples. Detection is colorimetric, based on TMB substrate development, and the assay is run on a pre-coated microplate. The kit is intended for research use, supporting the cell biology research area, where MCP4 — a C-C motif chemokine also known as CCL13, monocyte chemotactic protein 4, MCP-4, NCC-1, and small-inducible cytokine A13 — is studied for its role in recruiting immune cells to sites of inflammation and infection and in regulating immune cell migration and activation.

Analytical performance is characterised by a sensitivity of 9.375 pg/mL and a standard curve range of 15.625–1000 pg/mL. Linearity testing across serum, EDTA plasma, and heparin plasma (n = 5) is reported at 1:2, 1:4, and 1:8 dilutions, with overlapping limits at 1:2 of 88–97%, 89–104%, and 86–101% respectively, and proportionally similar values at higher dilutions. Spiked recovery across the same matrices averages 94% (serum), 96% (EDTA plasma), and 97% (heparin plasma), with range limits of 89–101%, 86–105%, and 85–105%. Sample preparation protocols are described for serum (clot, then centrifuge), plasma (anticoagulant tubes, centrifuged at 2–8 °C), tissue homogenate (PBS with protease inhibitors), cell culture supernatant (centrifuged at 2500 rpm for 5 minutes), and cell lysate (lysis buffer with protease inhibitors).

The assay procedure consists of reagent and plate equilibration, a primary incubation at 37 °C for 90 minutes, addition of a biotinylated detection antibody with a 60-minute incubation, HRP-streptavidin (SABC) binding for 30 minutes, TMB colour development in the dark for 10–20 minutes, and a stop step with absorbance read at 450 nm. Kit components include the pre-coated ELISA microplate (dismountable strips for sealed-foil storage), lyophilised standards, 100× biotin-labelled antibody, 100× HRP-streptavidin conjugate (SABC), TMB substrate, sample dilution buffer, antibody dilution buffer, SABC dilution buffer, stop solution, 25× wash buffer, plate sealers, and a technical manual. Storage recommendations are 2–8 °C for up to 12 months, with selected components also stable at –20 °C for extended periods where indicated.
