Description

From Assay Genie, this Sandwich ELISA kit uses a double-antibody configuration for the quantitative measurement of human MASP1 (Mannan-binding Lectin Serine Protease 1) in serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The colorimetric assay has been developed with high sensitivity and specificity for research applications relating to the lectin pathway of complement activation, innate immunity and inflammation, in which MASP1 is described as a pivotal protein.

Reported performance characteristics include a sensitivity of 56.25 pg/mL and a measuring range of 93.75–6000 pg/mL. Linearity in serum (n = 5) spans 93–105% at a 1:2 dilution, 92–98% at 1:4, and 93–101% at 1:8; in EDTA plasma (n = 5) the corresponding ranges are 83–99%, 89–101%, and 83–99%; and in heparin plasma (n = 5) they are 83–100%, 82–99%, and 90–100%. Spike-and-recovery averages are 95% for serum (range 86–103%), 96% for EDTA plasma (range 86–103%), and 94% for heparin plasma (range 85–102%).

The workflow consists of equilibrating reagents and TMB substrate to room temperature and loading standards, samples and blanks onto the pre-coated plate, followed by primary incubation at 37°C for 90 minutes, biotinylated detection antibody binding at 37°C for 60 minutes, HRP-Streptavidin (SABC) binding at 37°C for 30 minutes, TMB colour development in the dark for 10–20 minutes, and a stop reaction with absorbance read at 450 nm. Recommended storage is 2–8°C for 12 months. Sample preparation guidance includes allowing blood to clot and centrifuging at 1000 × g for 20 minutes for serum, using anticoagulant tubes and centrifuging at 1000 × g for 15 minutes at 2–8°C for plasma, and centrifuging cell culture supernatant at 2500 rpm for 5 minutes.
