Description

MAPKAPK5 (also called PRAK) is a 54 kDa serine/threonine kinase encoded on chromosome 12q24.13, catalogued at UniProt entry Q8IW41, that is activated downstream of p38α/β and ERK3/ERK4 and translocates between the nucleus and cytoplasm upon phosphorylation at Thr-182. The Assay Genie Human MAPKAPK5 ELISA Kit uses a quantitative double-antibody sandwich configuration on a pre-coated microplate, with streptavidin-HRP readout, biotinylated detector antibody, and TMB substrate chemistry. Each microplate provides 96 Assays and is compatible with standard plate-reader workflows. Reconstituted standards should be aliquoted to limit freeze-thaw cycles and stored refrigerated; protect reagents from light, and equilibrate all components to room temperature before opening. The kit measures endogenous human MAPKAPK5 with no significant cross-reactivity against homologous MAPKAPK family members such as MAPKAPK2 and MAPKAPK3, supporting studies of p38-MK5/PRAK-mediated phosphorylation of HSP27 at physiologically relevant sites, p53 activation at Ser37, oncogenic-RAS-induced senescence, and cytoskeletal remodeling via myosin light-chain regulation. Typical applications include biomarker profiling in serum and plasma from cohorts investigating tumor suppression, inflammatory signaling, and cellular stress responses. A 100 uL sample volume per well is typical, and absorbance values can be back-calculated against a recombinant human MAPKAPK5 standard curve for concentration determination. For research use only.
