A quantitative enzyme-linked immunosorbent (ELISA) assay from Assay Genie targets Mitogen-Activated Protein Kinase Kinase Kinase 5 (MAP4K5), the STE20-family serine/threonine kinase otherwise referenced in the literature as KHS or GCKR. MAP4K5 transmits extracellular stress cues — including TNF-alpha stimulation, oxidative challenge, and growth-factor engagement — through the MKK4/MKK7–JNK cascade, and is implicated in hepatocellular carcinoma proliferation, glucolipotoxicity-driven beta-cell apoptosis, and innate immune cytokine production in published studies.

Investigators profile endogenous MAP4K5 levels in human cell-line lysates and tissue homogenates alongside downstream phospho-JNK and phospho-c-Jun readouts to dissect kinase cascade wiring in oncology, metabolic-disease, and inflammation projects. The kit supports up to 96 assays on a single microplate when the recombinant standard curve is run in duplicate, and ships with pre-coated wells, biotinylated detection antibody, lyophilized protein standards, streptavidin-HRP conjugate, wash buffer concentrate, and TMB substrate.

Standard protocol steps — incubation, wash, chromogenic development, and stop-solution termination — produce an absorbance signal read on a benchtop plate reader. Reconstituted standards and detection antibody are kept refrigerated between runs and protected from light, while unused strip wells are returned to the foil pouch with desiccant to preserve activity through the printed expiration date.
