Description

A sandwich ELISA from Assay Genie is configured for the quantitative detection of MAP3K5/ASK1 in human samples, using a double-antibody format and a colorimetric read-out with absorbance measured at 450 nm [S1][C0]. Validated sample types include serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples [S1].

The kit has a reported sensitivity of 0.188 ng/mL and an assay range of 0.313–20 ng/mL [S1]. Linearity was evaluated in serum, EDTA plasma and heparin plasma at 1:2, 1:4 and 1:8 dilutions, with values of 94–105%, 82–98% and 82–96% for serum (n = 5), 89–103%, 86–100% and 80–96% for EDTA plasma (n = 5), and 85–97%, 90–101% and 82–97% for heparin plasma (n = 5) [S1]. Spike-and-recovery testing yielded ranges of 86–96% (average 91%) for serum, 85–99% (average 90%) for EDTA plasma, and 85–102% (average 94%) for heparin plasma (n = 5) [S1].

MAP3K5/ASK1 (UniProt Q99683) is positioned in the cell death research area, with reported roles in apoptosis, inflammation and oxidative stress pathways [S1]. The sample workflow includes a primary incubation at 37°C for 90 minutes, biotin-labelled detection antibody for 60 minutes, HRP-streptavidin (SABC) for 30 minutes, and TMB substrate development in the dark for 10–20 minutes before stop solution and absorbance reading [S1]. Kits are stored at 2-8°C for 12 months [S1].
