Description

A specialized sandwich ELISA assay from Assay Genie is designed for the accurate quantification of MAO-A (Monoamine Oxidase-A) levels in human samples, including serum, plasma, cell culture supernatants, cell or tissue lysates, and other liquid samples. MAO-A is an enzyme involved in the breakdown of neurotransmitters such as serotonin, dopamine, and norepinephrine, and dysregulation of its activity has been linked to psychiatric disorders including depression, anxiety, and schizophrenia.

The kit uses a double-antibody sandwich format with colorimetric detection. Its sensitivity is 9.375 pg/mL and the assay range is 15.625–1000 pg/mL. In linearity testing, serum (n = 5) showed 85–99%, 86–101% and 87–104% at 1:2, 1:4 and 1:8 dilutions; EDTA plasma (n = 5) showed 82–90%, 87–96% and 83–101%; and heparin plasma (n = 5) showed 84–91%, 84–98% and 81–100%. Recovery averaged 100% for serum (range 93–104%), 93% for EDTA plasma (range 87–100%) and 90% for heparin plasma (range 87–93%).

The protocol involves equilibrating reagents and TMB substrate to room temperature, a primary incubation at 37°C for 90 minutes, biotin-labeled detection antibody incubation at 37°C for 60 minutes, HRP-Streptavidin (SABC) binding at 37°C for 30 minutes, color development with TMB substrate in the dark for 10–20 minutes, and absorbance measurement at 450 nm after adding stop solution. Kit components include a dismountable ELISA microplate, lyophilized standard, biotin-labeled antibody (100X), HRP-Streptavidin conjugate SABC (100X), TMB substrate, sample dilution buffer, antibody dilution buffer, SABC dilution buffer, stop solution, 25X wash buffer, plate sealer and a technical manual. The kit should be stored at 2–8°C for 12 months.
