Beyond its role as a TNF-receptor superfamily member, LTbR (TNFRSF3) is the signalling receptor for the heterotrimeric lymphotoxin LTα1β2 and the TNF-family ligand LIGHT (TNFSF14). Ligation triggers the NIK-dependent non-canonical NF-κB pathway, driving RelB/p52 nuclear translocation and transcription of genes that orchestrate splenic white pulp organisation, Peyer's patch maintenance, and mucosal IgA responses.

Assay Genie supplies the kit with a lyophilised recombinant LTbR standard and a pre-titrated biotinylated detection antibody; the conjugate diluent and wash buffer are provided at 10× and 25× working concentrations, with TMB substrate and stop solution packaged separately to preserve chromogenic stability. Each lot ships with an assay-specific Certificate of Analysis reporting intra-assay precision CV below 8% and inter-assay CV below 10%.

Common research contexts include tracking soluble LTbR shedding in inflammatory bowel disease and rheumatoid arthritis synovial fluid, measuring stromal receptor expression in tumour xenograft lysates, and characterising LTβR signalling in primary hepatocyte and intestinal epithelial cultures. The capture antibody is human-specific, with no significant cross-reactivity reported for mouse or rat orthologues, which makes the assay directly applicable to xenograft and human primary cell studies. Bring all kit components to room temperature (18-25 °C) before use, and equilibrate the lyophilised standard by brief centrifugation before reconstitution with the supplied diluent to avoid pipetting loss.
