This in vitro immunoassay from Assay Genie quantitatively measures human Lymphocyte Antigen 9 (LY9), a SLAM-family surface receptor also known as CD229, in research samples. Validated for use with cell lysates and tissue homogenates, the kit delivers a colorimetric readout in a microplate layout compatible with standard laboratory plate readers. Each kit provides reagents for 96 determinations, including pre-coated wells, assay diluent, detection antibody, wash buffer, standards, and substrate solution. The protocol follows a sandwich-format procedure in which captured analyte is bound by a labeled secondary antibody, producing a chromogenic signal proportional to LY9 concentration in the loaded specimen.

LY9 (CD229/SLAMF3) is a homophilic adhesion molecule expressed on T lymphocytes, B cells, NK cells, and other immune cell populations. It modulates lymphocyte activation, cytokine production, and immunological synapse formation, making the marker a useful readout in research on autoimmune disease, lymphoid malignancies, transplant immunology, and broader immune signaling pathways. Laboratories studying chronic lymphocytic leukemia, multiple sclerosis, and adaptive immune regulation frequently use this receptor as a phenotyping or functional indicator.

Reagents should be stored refrigerated upon arrival, with light-sensitive components handled accordingly and all materials equilibrated to room temperature before each run. Repeated freeze-thaw cycles of the standards and detection antibody should be avoided. Components supplied are intended for research use only and have not been validated for diagnostic procedures. When handled per the supplied protocol, the kit retains performance through the expiration date printed on the outer label.
