Description

A colorimetric sandwich ELISA in a double-antibody format, manufactured by Assay Genie for the quantitative measurement of human LXR-alpha (also known as NR1H3, oxysterols receptor LXR-alpha, LXRA, LXR-a, or RLD-1). The target is a nuclear receptor that plays a critical role in regulating lipid metabolism and inflammation, and whose dysregulation has been implicated in atherosclerosis, metabolic disorders, and inflammatory conditions. The kit accepts serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples, and falls within the research area of epigenetics and nuclear signaling.

Reported analytical performance includes a sensitivity of 0.188 ng/mL and a detection range of 0.313–20 ng/mL, with absorbance read at 450 nm. Linearity (n=5) at dilutions 1:2, 1:4, and 1:8 spans 88–99%, 85–100%, and 88–103% for serum; 83–97%, 82–97%, and 83–101% for EDTA plasma; and 80–96%, 87–96%, and 84–98% for heparin plasma. Spike-and-recovery averages are 97% for serum (range 85–102%), 95% for EDTA plasma (range 88–102%), and 94% for heparin plasma (range 85–102%). Unopened components are stored at 2–8°C for up to 12 months, with several reagents marked as light-sensitive.

The supplied protocol describes reagent equilibration, a primary incubation at 37°C for 90 minutes, biotin-labeled detection antibody binding for 60 minutes, HRP-streptavidin (SABC) binding for 30 minutes, TMB substrate development in the dark for 10–20 minutes, and a stop reaction with absorbance reading at 450 nm. Sample preparation uses standard centrifugation protocols: serum is clotted and centrifuged at 1000 × g for 20 minutes; plasma is collected in anticoagulant tubes and centrifuged at 1000 × g for 15 minutes at 2–8°C; cell culture supernatant is centrifuged at 2500 rpm for 5 minutes; and tissue homogenates or cell lysates are prepared in PBS or lysis buffer with protease inhibitors.
