Human LOXL3/Lysyl Oxidase Homolog 3 ELISA Kit

Assay Genie's Human LOXL3/Lysyl Oxidase Homolog 3 ELISA Kit is a sandwich ELISA, double antibody immunoassay designed for the quantitative measurement of LOXL3 in human samples. The kit is reactive with human samples including serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples, and is supplied in a colorimetric format. The kit is intended for use within immunology research.

According to the manufacturer specifications, the assay has a sensitivity of 0.281 pmol/mL and a measuring range of 0.469–30 pmol/mL. Reported linearity for serum (n = 5) across 1:2, 1:4 and 1:8 dilutions is 82–100%, 81–100% and 90–98%, with EDTA plasma (n = 5) at 82–98%, 82–92% and 85–105%, and heparin plasma (n = 5) at 85–100%, 87–101% and 82–99%. Spike-and-recovery performance for serum (n = 5) is 87–103% (average 95%), EDTA plasma (n = 5) 85–100% (average 93%), and heparin plasma (n = 5) 85–105% (average 90%). Storage is recommended at 2–8 °C for 12 months.

The assay procedure involves equilibrating reagents and TMB substrate to room temperature, loading standards, samples and a zero control onto the pre-coated plate and incubating at 37 °C for 90 minutes, binding the biotin-labelled detection antibody at 37 °C for 60 minutes, adding HRP-Streptavidin (SABC) for 30 minutes at 37 °C, developing with TMB substrate for 10–20 minutes in the dark, and reading absorbance at 450 nm after adding stop solution. LOXL3 is described as an enzyme involved in extracellular matrix remodelling and cross-linking of collagen and elastin, with reported associations with fibrosis, cancer and connective tissue disorders.
