Description
Manufactured by Assay Genie, this Sandwich ELISA kit enables quantitative detection of human DNA Ligase 3 (LIG3) in research samples. It uses a double-antibody sandwich format with colorimetric readout and accepts serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The assay has a sensitivity of 0.188 ng/mL and a working range of 0.313–20 ng/mL, and the kit is intended for storage at 2–8°C for up to 12 months.
Linearity was validated across serum, EDTA plasma, and heparin plasma (n = 5 each), and spike-recovery testing returned averages of 96% for serum, 99% for EDTA plasma, and 95% for heparin plasma. The sample protocol proceeds through reagent equilibration at room temperature, a 37°C primary incubation of 90 minutes, biotinylated detection antibody binding for 60 minutes, HRP-streptavidin binding for 30 minutes, TMB substrate colour development in the dark for 10–20 minutes, and absorbance reading at 450 nm after stop solution addition.
DNA Ligase 3 is an ATP-dependent enzyme involved in DNA repair and replication, contributing to the maintenance of genomic stability. Dysregulation of the enzyme has been linked to cancer, genetic disorders, and neurodegenerative conditions. The kit is annotated for the Cell Cycle research area, with the target protein carrying UniProt identifier P49916 and the synonyms LIG3, DNA ligase III, and Polydeoxyribonucleotide synthase [ATP] 3.
