Human LIF/Leukemia inhibitory factor ELISA Kit

This Sandwich ELISA Kit from Assay Genie is designed for the quantitative measurement of Leukemia Inhibitory Factor (LIF) in human samples, including serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Using a double antibody sandwich format with colorimetric detection, the kit targets a cytokine involved in cell growth regulation, immune response, inflammation, and embryonic development. LIF is described as a valuable biomarker relevant to conditions such as leukemia, cancer, and inflammatory disorders.

The assay has a sensitivity of 18.75 pg/mL and a working range of 31.25–2000 pg/mL (1 ng = 12700 mIU). Reported linearity in serum spans 87–103%, 85–105%, and 80–99% across serial dilutions; EDTA plasma linearity spans 86–98%, 89–101%, and 82–100%; and heparin plasma spans 82–98%, 86–101%, and 85–96%. Spike-and-recovery averages are 96% for serum, 94% for EDTA plasma, and 95% for heparin plasma.

The workflow involves equilibrating reagents, incubating standards and samples on a pre-coated plate, adding biotin-labeled detection antibody, binding HRP-Streptavidin, developing with TMB substrate in the dark, and reading absorbance at 450 nm after stopping the reaction. Kit components include a dismountable ELISA microplate, lyophilized standard, biotin-labeled antibody, HRP-Streptavidin conjugate, TMB substrate, sample and antibody dilution buffers, SABC dilution buffer, stop solution, wash buffer, plate sealer, and a technical manual. Components should be stored at 2–8°C, with the microplate and standard also suitable for longer-term storage at -20°C.
