Human Leukosialin/SPN ELISA Kit

Manufactured by Assay Genie, this sandwich ELISA kit uses a double-antibody format for the quantitative measurement of human Leukosialin (SPN), a cell surface glycoprotein also known as CD43, Sialophorin and Galactoglycoprotein (UniProt P16150). It is validated for serum, plasma, cell culture supernatant, cell or tissue lysate and other liquid samples, and should be stored at 2–8°C for up to 12 months.

Linearity has been demonstrated across dilution ratios of 1:2, 1:4 and 1:8: serum (n=5) at 88–102%, 90–102% and 86–104%; EDTA plasma (n=5) at 87–100%, 85–93% and 87–99%; heparin plasma (n=5) at 80–92%, 88–96% and 87–100%. Spike-and-recovery averages are 98% for serum (range 95–102%), 96% for EDTA plasma (range 94–100%) and 92% for heparin plasma (range 86–101%).

The workflow proceeds through reagent equilibration, primary incubation at 37°C for 90 minutes, biotin-labelled detection antibody binding for 60 minutes, HRP-Streptavidin incubation for 30 minutes, TMB colour development in the dark for 10–20 minutes, and a stop reaction with absorbance read at 450 nm. Sample-preparation protocols are supplied for serum, plasma, tissue homogenate, cell-culture supernatant and cell lysate.
