Human Leptin ELISA Kit

Manufactured by Assay Genie, this sandwich double-antibody ELISA is designed for the quantitative measurement of human Leptin in serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The microplate supplied is pre-coated with a capture antibody specific to the target protein. Standards and appropriately diluted samples are added to the wells and incubated, allowing the target analyte to bind to the immobilised antibody. A biotinylated detection antibody directed at a different epitope is then added to form a sandwich complex, after which HRP-conjugated Streptavidin and TMB substrate generate a colorimetric reaction. The reaction is terminated with an acidic stop solution and absorbance is read at 450 nm using a microplate reader. Signal intensity is directly proportional to analyte concentration and is determined against a standard curve.

The kit has a sensitivity of 18.75 pg/mL and a measuring range of 31.25–2000 pg/mL (1 ng = 1200 mIU). Linearity in serum (n = 5) ranges from 85–93% at 1:2, 82–100% at 1:4 and 85–97% at 1:8; in EDTA plasma from 90–104%, 87–99% and 85–91%; and in heparin plasma from 88–92%, 86–101% and 80–98%. Recovery values are 85–98% (average 91%) for serum, 86–99% (average 92%) for EDTA plasma and 88–102% (average 95%) for heparin plasma. All reagents should be stored at 2–8°C and remain stable for 12 months under those conditions.
