The Assay Genie kit enables quantitative measurement of LEMD3, an integral inner nuclear membrane protein encoded by the LEMD3 gene that anchors chromatin to the nuclear envelope via interaction with A-type lamins and barrier-to-autointegration factor (BAF). Beyond its structural role at the nuclear periphery, LEMD3 antagonizes BMP and TGF-β signaling by sequestering receptor-activated SMAD proteins in the nucleus, a function relevant to bone and connective tissue biology. Loss-of-function mutations in LEMD3 are linked to Buschke-Ollendorff syndrome and melorheostosis, making accurate quantification of interest in musculoskeletal, dermatological, and cell-signaling research. Reagents are supplied in sealed vials and pre-coated microplate wells; reconstituted standards and biotinylated detection antibody should be stored at -20°C and protected from light, with limited freeze-thaw cycles recommended to preserve binding activity.
