The Assay Genie sandwich ELISA workflow uses a biotinylated detection antibody paired with an Avidin-Horseradish Peroxidase (HRP) conjugate. After sample incubation, TMB substrate is added to develop a blue colorimetric signal that turns yellow upon addition of the stop solution, with the optical density read at 450 nm. Standards are run in parallel to generate a calibration curve from which sample concentrations are interpolated.

Typical run parameters use 100 µL of sample or standard per well, with incubation at 37 °C across the sequential binding, HRP, and substrate steps. The complete assay procedure is generally completed in approximately 3 hours. Reported precision is intra-assay CV below 8% and inter-assay CV below 10%, and the kit shows minimal cross-reactivity with related thyroid autoantibody analytes, supporting selective measurement of LATS activity in complex biological matrices.

Reagent components include a pre-coated 96-well microplate, biotinylated antibody working solution, HRP-avidin conjugate, TMB substrate, stop solution, wash buffer concentrate (typically 25X), and a serial dilution standard set. Unopened kits should be stored at 2-8 °C and protected from light, where they remain stable for up to 12 months; reconstituted or working solutions may be aliquoted and held at -20 °C for longer-term use. Bring all reagents to room temperature before initiating the assay, and avoid repeated freeze-thaw cycles of prepared working solutions to preserve antibody and conjugate activity.
