Description

A Sandwich ELISA kit from Assay Genie designed for the quantitative measurement of human LAT (Linker for Activation of T Cells) using a double-antibody, colorimetric workflow. The assay is intended for human samples and is validated for use with serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Reagents should be stored at 2-8°C, with a shelf life of up to 12 months.

According to the manufacturer's specifications, the kit has a sensitivity of 0.188 ng/mL and a working range of 0.313-20 ng/mL. The procedure involves a pre-coated capture plate, incubation with sample and standards at 37°C for 90 minutes, biotin-labelled detection antibody for 60 minutes, HRP-Streptavidin (SABC) for 30 minutes, and TMB substrate development in the dark for 10-20 minutes, with absorbance read at 450 nm. Reported linearity (1:2, 1:4, 1:8 dilutions; n = 5) is 85-104% for serum, 87-95% for EDTA plasma, and 88-104% for heparin plasma, with average recovery of 96% for serum, 98% for EDTA plasma, and 98% for heparin plasma.

LAT is a signalling molecule involved in T cell activation and immune response regulation, and its dysregulation has been linked to autoimmune diseases, allergies, and immune deficiencies. The kit is supplied as research-use-only reagents with components including a dismountable ELISA microplate, lyophilised standard, biotin-labelled antibody concentrate, HRP-Streptavidin conjugate, TMB substrate, sample and antibody dilution buffers, wash buffer, stop solution, and plate sealer.
