This quantitative immunoassay from Assay Genie measures Lysosomal-Associated Membrane Protein 2 (LAMP2), a heavily glycosylated single-pass transmembrane glycoprotein of roughly 95-120 kDa on immunoblots, also known as CD107b. LAMP2 is predominantly localized to the limiting membrane of late endosomes and lysosomes, where isoform B supports chaperone-mediated autophagy by acting as the substrate receptor for cytosolic proteins carrying a KFERQ-like targeting motif. Loss-of-function mutations in the LAMP2 gene cause Danon disease, an X-linked lysosomal storage disorder characterized by hypertrophic cardiomyopathy, skeletal myopathy, and variable intellectual disability, which is why circulating LAMP2 levels are studied as a biomarker in cardiovascular and metabolic disease research. CD107b is also widely cited as a cell-surface degranulation marker on cytotoxic lymphocytes, including NK cells and activated CD8+ T cells, supporting immunology, cell biology, and inflammation studies. The kit uses a human-specific capture antibody paired with a matched detection reagent in a microplate format and has not been validated against rodent orthologues, so it functions as a human-only measurement. Reconstituted standards and prepared samples are handled according to the lot-specific Certificate of Analysis supplied with each vial set, and unopened components are stored refrigerated between uses to preserve antibody binding activity. Applications include quantifying LAMP2 release into cell culture supernatants, profiling plasma from cardiomyopathy cohorts, and screening serum for lysosomal membrane integrity changes during drug-response studies.
