Description

A sandwich ELISA kit from Assay Genie designed for the quantitative measurement of human L-Selectin/CD62L, a cell adhesion molecule involved in leukocyte rolling and the recruitment of immune cells to sites of inflammation. The assay uses a double-antibody sandwich format with colorimetric detection and is intended for human serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples.

Performance characteristics include a sensitivity of 46.875 pg/mL and a working range of 78.125-5000 pg/mL. Linearity in serum (n=5) is reported as 94-105% at 1:2, 92-101% at 1:4, and 85-98% at 1:8; EDTA plasma (n=5) at 95-99%, 83-94%, and 83-100%; and heparin plasma (n=5) at 81-98%, 81-99%, and 80-97%. Recovery ranges are 86-105% (average 95%) for serum, 87-105% (average 94%) for EDTA plasma, and 89-102% (average 97%) for heparin plasma.

The assay procedure follows six steps: reagent and plate preparation, primary incubation at 37°C for 90 minutes, detection antibody binding for 60 minutes, HRP-streptavidin binding for 30 minutes, TMB color development in the dark for 10-20 minutes, and stop reaction with absorbance measurement at 450 nm. The kit is stored at 2-8°C for 12 months and is relevant to cell biology research, where dysregulation of L-Selectin has been implicated in inflammatory and autoimmune disorders.
