Human Klotho ELISA Kit

A sandwich ELISA from Assay Genie designed for the measurement of human Klotho in biological samples. Klotho is a protein implicated in the regulation of aging and age-related processes, including metabolism, oxidative stress, and growth factor signalling, and its dysregulation has been linked to a range of health conditions. The assay is intended for research applications in cell biology and the study of aging and age-related disorders.

The kit has a stated sensitivity of 4.688 pg/mL and an assay range of 7.813–500 pg/mL. It is reactive with human samples and has been validated for serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Storage is at 2–8°C for 12 months. Reported linearity values are 86–104%, 82–101%, and 82–100% for serum at 1:2, 1:4, and 1:8 dilutions respectively; EDTA plasma ranges from 82–105% and heparin plasma from 80–105% across the same dilutions. Recovery averages are 94% for serum, 99% for EDTA plasma, and 90% for heparin plasma.

The procedure follows a sandwich ELISA, double-antibody format using a biotin-labelled detection antibody and HRP-streptavidin (SABC). Standards, samples, and blanks are incubated on the pre-coated plate at 37°C for 90 minutes, followed by detection antibody incubation at 37°C for 60 minutes, HRP-streptavidin binding for 30 minutes, TMB substrate development in the dark for 10–20 minutes, and absorbance reading at 450 nm after stop solution addition.
